A novel organocatalytic reaction simplifies the creation of oligonucleotide drugs based on modified nucleic acid chains. Ming Shang and colleagues at Shanghai Jiao Tong University in China achieved this by replacing temporary molecular guides. The method was demonstrated across over 20 nucleoside combinations, successfully forming various phosphorus bonds. This approach offers a less expensive and scalable alternative to older methods that required bulky chiral auxiliaries. Ultimately, this research aims to better understand how phosphorus stereochemistry impacts drug performance. π§¬
